METHODOLOGICAL INFORMATION
Description of methods used for collection/generation of data: Fluorescence measurements at different wavelengths provide quantitative and qualitative data about intracellular molecules such as DNA. Once the optics directs the light to a detector, the electronics system turns that light into an electronic signal. Each signal pulse represents an individual event (or cell). Cytometers display data in terms of height, width, and area, describing the properties of the signal pulse.
Methods for processing the data: BD FACSDiva 8.0.1 software offers efficient data management tools to simplify data storage and improve flow cytometer data organization. It provides automated data export in XML, FCS, and even ZIP formats. Manual export of FCS files creates an experiment folder structure in the operating system for easy identification of data files without the need to open them. While FCS files are widely used, they can be challenging due to their proprietary format. For that reason, they were converted to PDF.
Instrument- or software-specific information needed to interpret the data: BD FACSDiva™ Software 8.0.1 provides a wide array of basic analysis features enabling multiple applications to run on the BD FACSCanto Flow Cytometers.
Standards and calibration information, if appropriate: Cytometer settings are tracked over time by Levey-Jennings graphs to monitor cytometer performance. Adjustments are made automatically to ensure consistent data integrity and reproducible results from day to day and experiment to experiment.
Environmental/experimental conditions: Measurements were performed at standard conditions.
Describe any quality-assurance procedures performed on the data: Integrated quality control features further support data integrity by notifying operators if an assay fails predefined standards.
